Next PageAbstract: | Seven hundred and thirty fishes and 276 crustaceans collected from various fish markets of Coimbatore, South India, over a period of 2 years (September 1990 to August 1992) were analysed for the prevalence of Salmonella. Fishes (14·25%) and 17·39% of crustaceans were found to be contaminated with Salmonella. Of the different fishes analysed, the highest incidence of Salmonella was seen in Scopelidae (28%) followed by Trachnidae (26·9%). Among crustaceans Portunus pelagicus (33·33%) showed the highest incidence followed by Scylla serrata (28·57%). A well-marked seasonal variation in the incidence pattern was observed in both fishes and crustaceans with a higher incidence during monsoon season followed by post-monsoon and pre-monsoon. The region of the body that showed frequent isolation was the alimentary canal in fishes (41·33%) and gills (35·06%) in crustaceans. Serotyping of the isolates revealed prevalence of Salmonella weltevreden, Salmonella typhi, Salmonella paratyphi B, Salmonella mgulani and Salmonella typhimurium in both fishes and crustaceans. Abstract: | One thousand, two hundred and sixty four samples of individually quick-frozen (IQF) peeled and deveined raw and 914 samples of cooked ready to eat shrimp samples produced from farm raised black tiger (Penaeus monodon) obtained from a seafood unit working under HACCP concept were analysed for total aerobic plate count (APC), coliform count, Escherichia coli, coagulase positive Staphylococci and Salmonella. The overall bacteriological quality of the product was found to be good. Of the frozen raw shrimp, 96% of samples showed APC below 105 while 99% of the frozen cooked ready-to-eat samples showed APC less than 104. The APC ranged from 1·0´102 to 4·2´106 cfu/gm in frozen raw shrimp and from 1·0´102 to 6·4´104 cfu/gm in the frozen cooked shrimp. Prevalences of coliforms in raw shrimp and cooked shrimp samples were 14·4% and 2·9% respectively. The coliform count in raw products ranged from 1·0´101 to 2·5´103 cfu/gm and in the cooked products, from 1·0 ´101 to 1·8´102 cfu/gm. Although all the cooked shrimp samples were free of coagulase positive staphylococci, E. coli and Salmonella, 1·0, 2·0 and 0·1% of the frozen raw shrimp samples tested positive for coagulase positive Staphylococci, E. coli and Salmonella respectively. The Salmonella strain was identified as Salmonella typhimurium. The results of the present study highlight the importance of implementation of HACCP system in the seafood industry to ensure consistent quality of frozen seafood |

Abstract: | A total of 319 strains of Aeromonas hydrophila were isolated from 536 fish and 278 prawns for a 2-year period. All the strains were tested for resistance to 15 antibiotics and 100% of the strains was resistant to methicillin and rifampicin followed by bacitracin and novobiocin (99%). Only 3% of the strains exhibited resistance against chloramphenicol. Abstract: | Bacteriological quality of individually quick frozen (IQF) shrimp products produced from aquacultured tiger shrimp (Penaeus monodon) has been analysed in terms of aerobic plate count (APC), coliforms, Escherichia coli, coagulase-positive staphylococci, Salmonella, and Listeria monocytogenes. Eight hundred forty-six samples of raw, peeled, and deveined tail-on (RPTO), 928 samples of cooked, peeled, and deveined tail-on (CPTO), 295 samples of headless, undeveined shell-on (HLSO), and 141 samples of raw, peeled, and deveined tail-off (RPND) shrimps were analysed for the above bacteriological parameters. Salmonella was isolated in only one sample of raw, peeled tail-on. Serotyping of the strain revealed that it was S. typhimurium. While none of the cooked, peeled tail-on shrimp samples exceeded the aerobic plate count (APC) of 105 colony forming units per gram (cfu/g), 2.5% of raw, peeled, tail-on, 6.4% of raw, peeled tail-off, and 7.5% of headless shell-on shrimp samples exceeded that level. Coliforms were detected in all the products, though at a low level. Prevalence of coliforms was higher in headless shell-on (26%) shrimps followed by raw, peeled, and deveined tail-off (19%), raw, peeled tail-on (10%), and cooked, peeled tail-on (3.8%) shrimps. While none of the cooked, peeled tail-on shrimp samples were positive for coagulase-positive staphylococci and E. coli, 0.6–1.3% of the raw, peeled tail-on were positive for staphylococci and E. coli, respectively. Prevalence of staphylococci was highest in raw, peeled tail-off (5%) shrimps and the highest prevalence of E. coli (4.8%) was noticed in headless shell-on shrimps. L. monocytogenes was not detected in any of the cooked, peeled tail-on shrimps. Overall results revealed that the plant under investigation had exerted good process control in order to maintain superior bacteriological quality of their products |

Abstract: | The survival of Escherichia coli in tropical estuarine water has been studied under controlled laboratory conditions using microcosms. The survival has been assessed in terms of various self purifying factors of the natural waters such as biological, chemical and physical factors. The biological factors considered included competition from other microorganisms, predation by protozoa and coliphages. The suitability of the chemical composition of estuarine water has been studied under chemical factors and negative impact of sunlight has been studied under physical factors. The results revealed that sunlight exerted maximum negative impact, followed by biotic factors contained in the estuarine water. However, the chemical composition of the estuarine water is found to be suitable for the growth and survival of E. coli. The injury exerted by each of the above factors was also evaluated by using a selective and non-selective medium in conjunction. It was found that sunlight resulted in 100% injury of the cells as the cells failed to develop in a selective medium. While, sunlight resulted in the extinction of 90% of the E. coli cells within the first two hours of exposure, biotic factors took nearly 24 hours to remove the same amount of population. |

Abstract: | Prevalence and antibiotic resistance of Escherichia coli in the water and sediment samples of brackish water aquaculture ponds adjacent to Cochin backwaters was analysed. More than 50% of the water samples and more than 80% of sediment samples from all the sampling stations were tested positive for £. coli. Risk assessment of the E. coli strains was carried out using multiple antibiotic resistance (MAR) indexing. Majority of the strains were found to be multiple antibiotic resistant suggesting their origin from high risk sources of contamination such as human where antibiotics are frequently used. While none of the £. coli strains were resistant against amikacin, chloramphenicol, streptomycin and trimethoprim, considerable levels of resistance was encountered against ampicillin, erythromycin, penicillin G and vancomycin. High prevalence of £. coli in the water and sediment samples of this extensive brackish water ponds indicates high degree of faecal pollution of this environment. The high risk nature of the strains warrants efficient post harvest and processing measures to avoid health risk to consumers |

Abstract: | Five hundred and thirty six samples offishes and 278 prawn samples from the major fish market ofCoimbatore, South India, were analysed for the prevalence of Aeromonas hydrophila over a period of2 years (June 1997–May 1999). The prevalence level of A. hydrophila varied from 17.62% in prawns to 33.58% in fishes. More than 30% of the popular table fishes such as Sardinella longiceps, Rastrelliger kanagurta, Mugil cephalus and Caranx sexfasciatus were tested positive for this organism. Among the different species of the prawns analysed, Penaeus semisulcatus showed higher incidence (23.52%). Seasonal variation in the prevalence levels of A. hydrophila in fish and prawns revealed a higher prevalence during the monsoon season during 1997–98 and 1998–99. Of the different body parts of the fishes analysed for A. hydrophila, the intestinal samples showed higher prevalence (38.43%), followed by body surface (32.46%) and gill (29.10%). Considering the psychrotrophic nature and role of A. hydrophila as a pathogen ofemerging importance, the considerably high levels ofthis organism in a popular food item such as fish and prawn raises serious concern |

Abstract: | Motile aeromonads isolated from the intestines of farm-raised freshwater fish such as Catla catla, Labeo rohita and Ctenopharyngodon idella have been characterized to species level. Morphological and physiological grouping revealed 61% Aeromonas hydrophila, 30% Aeromonas caviae, 7% Aeromonas sobria and 2% which remained unidentified. Hemolytic activity was detected mostly in A. hydrophila, while only half of the A. sobria and A. caviae showed this activity. Antibiotic resistance patterns of the strains revealed that they had acquired a relatively higher resistance to oxytetracycline, amoxycillin, ampicillin, novobiocin and polymixin-B, implicating possible use of these antibiotics in the aquaculture systems. |

Abstract: | Microcosm studies have been carried out to find out the relative survival of Escherichia coli and Salmonella typhimurium in a tropical estuary. Survival has been assessed in relation to the important self-purifying parameters such as biotic factors contained in the estuarine water, toxicity due to the dissolved organic and antibiotic substances in the water and the sunlight. The results revealed that sunlight is the most important inactivating factor on the survival of E. coli and S. typhimurium in the estuarine water. While the biological factors contained in the estuarine water such as protozoans and bacteriophages also exerted considerable inactivation of these organisms, the composition of the water with all its dissolved organic and inorganic substances was not damaging to the test organisms. Results also indicated better survival capacity of E. coli cells under all test conditions when compared to S. typhimurium |

A total of 492 eggs and 82 egg-storing trays were examined over a period of 1 year from different retail outlets of a residential area of Coimbatore city, South India. Salmonella contamination was recorded in 38 of 492 (7.7%) eggs out of which 29 was in eggshell (5.9%) and 9 in egg contents (1.8%). Around 7.5% of the egg-storing trays were also found to be contaminated with Salmonella. Serotyping of the Salmonella strains showed that 89.7% of the strains from eggshell, 100% of the strains from egg contents and 71.4% of the strains from egg-storing trays were Salmonella Enteritidis. Other serovarvars encountered were S. Cerro, S. Molade and S. Mbandaka from eggshell and S. Cerro from egg-storing trays. Seasonal variations in the prevalence pattern were identified with, a higher prevalence during monsoon months followed by post-monsoon and premonsoon. Further examination of the Salmonella strains was carried out by testing their antimicrobial sensitivity against 10 commonly used antimicrobials. Results revealed high prevalence of multiple antimicrobial resistance among these strains suggesting possible prior selection by use of antimicrobials in egg production |

Abstract: | The diversity and load of heterotrophic bacteria and fungi associated with the mangrove soil from Suva, Fiji Islands, was determined by using the plate count method. The ability of the bacterial isolates to produce various hydrolytic enzymes such as amylase, gelatinase and lipase were determined using the plate assay. The heterotrophic bacterial load was considerably higher than the fungal load. There was a predominance of the gram positive genus, Bacillus. Other genera encountered included Staphylococcus, Micrococcus, Listeria and Vibrio. Their effectiveness on the degradation of commercial polythene carry bags made of high density polyethylene (HDPE) and low density polyethylene (LDPE) was studied over a period of eight weeks in the laboratory. Biodegradation was measured in terms of mean weight loss, which was nearly 5 % after a period of eight weeks. There was a significant increase in the bacterial load of the soil attached to class 2 (HDPE) polythene. After eight weeks of submergence in mangrove soil, soil attached to class 1 and class 3 polythene mostly had Bacillus (Staphylococcus predominated in class 2 polythene). While most of the isolates were capable of producing hydrolytic enzymes such as amylase and gelatinase, lipolytic activity was low. Class 2 HDPE suffered the greatest biodegradation. |

Abstract: | A total of eighty-one Escherichia coli isolates belonging to forty-three different serotypes including several pathogenic strains such as enterotoxigenic E. coli (ETEC), enterohaemorrhagic E. coli (EHEC), enteropathogenic E. coli (EPEC) and uropathogenic E. coli (UPEC) isolated from Cochin estuary between November 2001 and October 2002 were tested against twelve antibiotics to determine the prevalence of multiple antibiotic resistance (MAR) and antimicrobial resistance profiles as a measure of high risk source of contamination. The results revealed that more than 95% of the isolates were multiple antibiotic resistant (resistant to more than three antibiotics). The MAR indexing of the isolates showed that all these strains originated from high risk source of contamination. The incidence of multiple antibiotic resistant E. coli especially the pathogenic strains in natural waters will pose a serious threat to human population |

Abstract: | Biosurfactants are surface active compounds released by microorganisms. They are biodegradable non-toxic and eco-friendly materials. In this review we have updated the information about different microbial surfactants. The biosurfactant production depends on the fermentation conditions, environmental factors and nutrient availability. The extraction of the biosurfactants from the cell-free supernatant using the solvent extraction procedure and the qualitative and quantitative analysis has been discussed with appropriate equipment details. The application of the biosurfactant includes biomedical, cosmetic and bioremediation. The type of microbial biosurfactants include trehalose lipids, rhamnolipids, sophorolipids, glycolipids, cellobiose lipids, polyol lipids, diglycosyl diglycerides, lipoloysaccharides, arthrofactin, lichensyn A and B, surfactin, viscosin, phospholipids, sulphonyl lipids and fatty acids. Rhamnolipid biosurfactants produced by Pseudomonas aeruginosa DS10-129 showed significant applications in the bioremediation of hydrocarbons in gasoline spilled soil and petroleum oily sludge. Rhamnolipid biosurfactant enhanced the bioremediation process by releasing the weathered oil from the soil matrices and enhanced the bioavailability of hydrocarbons for microbial degradation. It is having potential applications in the remediation of hydrocarbon contaminated sites. Biosurfactants from marine microorganisms also offer great potential in bioremediation of oil contaminated oceanic environments |

Abstract: | During an interstitial faunal survey along the south-west coast of Kerala, India, gastrotrich fauna were found in abundance. Together with species of the genera Xenotrichula, Halichaetonotus and Tetranchyroderma, were present several undescribed thaumastodermatid gastrotrichs belonging to the buccal palp bearing genus Pseudostomella. Adults of the new species are characterized by the following traits: total body length of about 300 μm; cuticular armature made up of medium sized pentancres covering the entire dorsolateral surface; pre-buccal, grasping palps bearing five, large papillae dorsally and 4-6 smaller papillae ventrally; adhesive apparatus made up of six anterior, 22-24 ventrolateral, two dorsolateral and six posterior adhesive tubes; caudal organ pear-shaped; frontal organ spherical. Pseudostomella cheraensis sp. nov. is the fourth taxon of the genus known from India; however, all the previous species reported hitherto from India have tetrancres instead of pentancres. |

Abstract: | Prevalence of faecal indicator bacteria, Escherichia coli and pathogenic bacteria, Vibrio cholerae, Vibrio parahaemolyticus and Salmonella were analysed in Vembanadu lake (98350N 768250E), along south west coast of India for a period of one year from ten stations on the southern and northern sides of a salt water regulator constructed in Vembanadu Lake in order to prevent incursion of seawater during certain periods of the year. While the northern side of the lake has a connection to the sea, the southern side is enclosed when the salt water regulator is closed. The results revealed the water body is polluted with high faecal coliform bacteria with mean MPN value ranging from 1718-7706/100 ml. E. coli, V. cholerae, V. parahaemolyticus and Salmonella serotypes such as S. paratyphi A, B, C and S. newport were isolated and this is the first report on the isolation of these Salmonella serovars from this lake. E. coli showed highest percentage of incidence (85.6–86.7%) followed by Salmonella (42–57%), V. choleare (40–45%) and V. parahaemolyticus (31.5–32%). The increased prevalence of indicator and pathogenic bacteria in the enclosed southern part of Vembanadu Lake may be resulting from the altered flow patterns due to the salt water regulator. |

Abstract: | Eight hundred and eighty-¢ve strains of bacterial isolates fromvarious samples associatedwith the natural habitat ofMacrobrachiumrosenbergii were screened for their probiotic potential. Two putative probionts namely Bacillus NL110 and Vibrio NE17 isolated from the larvae and egg samples, respectively, were selected for experimental studies and were introduced to the juveniles of M. rosenbergii (0.080 0.001g) through di¡erent modes such as through feed, water and both. The probiotic potential of the above bacteria in terms of improvements inwater quality, growth, survival, speci¢c growth rate (SGR), feed conversion ratio and immune parameters was evaluated. The treatment groups showed a signi¢cant improvement in SGR and weight gain (Po0.001). Survival among di¡erent treatment groups was better than that in the control group. There were also signi¢cant improvements in the water quality parameters such as the concentration of nitrate and ammonia in the treatment groups (Po0.05). Improvements in immune parameters such as the total haemocyte count (Po0.05), phenoloxidase activity and respiratory burst were also signi¢cant (Po0.001). It is concluded that screening of the natural micro£ora of cultured ¢sh and shell¢sh for putative probionts might yield probiotic strains of bacteria that could be utilized for an environment-friendly and organic mode of aquaculture. |
