Abstract: | Even though Bergey '5 Manual has been recognized globally as the guide to bacterial systematics, it has to be emphasized that descriptions given to a large extent are based on studies made with temperate isolates This leads one to conclude that any attempt to identify the tropical isolates with identification keys and tables generated from this information may lead to erroneous conclusions. And there is every possibility of the existence of genotypic and phenotypic variants or even nev. species in this part ofthe aquatic ecosystem. Applications ofa polythetic scheme of classification based on the principles of Numerical Taxonomy opens up exciting avenues for bringing to light, this possibility which otherwise would have been masked by the unidirectional approach as in monothetic schemes. Another added advantage of clustering a ‘natural’ bacterial population by numerical taxonomy, is the ease by which genotypic characterization could be performed on the clusters by selecting a representative from each cluster This helps overcome the practical impossibility of analyzing all the isolates in a pani:'_lar cluster. The genotypic characteizarion would either be mole °/o G-'rC. DNA-D.\_-X hybridization, DNA-RNA hybridization or DNA fingerprinting. Considering the requirement creating a broad base in the understanding of the family Vibrionaceae associated with the larvae ofM rosenbergii, the present work was undertaken to channelize every new information generated for developing appropriate managerial measures to protect the larvae from vibriosis during the unusually prolonged larval phase. |

Abstract: | Instead of developing easily degradable, and low-priced insecticides, we are going after highly sophisticated chemicals. Here, an attempt is being made to develop safer formulations of insecticides of botanical origin. Different parts of the plants were chosen based on their use in countryside and villages The dried plant materials were extracted with petroleum ether, and were applied on Tribolium castaneum. The results were statistically analysed. The active principles from Croton tigilium and Leea sambucina, the most potential plants, were isolated using Column Chromatography, TLC, and Hydrolysis. The isolated principles were analysed spectroscopically ( UV-Vis., IR, NMR, and MS ) to identify their chemical nature. The active principles from Leea and Croton were identified as a cholisterate derivative and a phorbol derivative respectively. In order to ascertain the environmental combatibility of the principles, degradation by soil bacteria was studied. The isolated principles were made into three type of formulations using stabilizers .The formulations were applied on Snake gourd semilooper, Pulse beetle, and mosquito larvae. Also the biocidal activity of the formulations was studied. Both Leea derivative and Croton derivative could be formulated effectively and were effective against a variety of pests. They are eco-friendly, as there is no artificial chemicals involved. |

Abstract: | The present work deals with the development of primary cell culture and diploid cell lines from two fishes, such as Poecilia reticulata and Clarias gariepinus. The greatest difficulty experienced was the avoidance of bacterial and fungi contamination. Three types of cell cultures are commonly developed, primary cell culture, diploid cell lines and heteroploid cell lines. Primary cell culture obtained from the animal tissues that have been cultivated in vitro for the first time. They are characterized by the same chromosome number as parent tissue, cultivated in vitro for the first time, have wide range of virus susceptibility, usually not malignant, six chromatin retarded and do not grow as suspension cultures. Diploid cell lines arise from a primary cell culture at the time of subculturing. Diploid cell lines commercially used in virology are W1-38 (human embryonic lung), W1-26 (human embryonic lung) and HEX (Human embryonic kidney). Heteroploid cell lines have been subcultivated with less than 75% of the cells in the population having a diploid chromosome constitution. Tissue cultures have been extensively used in biomedical research. The main applications are in three areas, Karyological studies, Identification and study of hereditary metabolic disorders and Somatic cell genetics. Other applications are in virology and host-parasite relationships. In this study an attempt was made to preserve the ovarian tissue at low temperature in the presence of cryoprotectants so that the tissue can be retrieved at any time and a cell culture could be developed. |

Abstract: | In the present study the development of bioreactors for nitrifying water in closed system hatcheries of penaeid and non-penaeid prawns. This work is an attempt in this direction to cater to the needs of aquaculture industry for treatment and remediation of ammonia and nitrate in penaeid and non-penaeid hatcheries, by developing nitrifying bacteria allochthonous to the particular environment under consideration, and immobilizing them on an appropriately designed support materials configured as reactors. Ammonia toxicity is the major limiting factors in penaeid and non-penaeid hatchery systems causing lethal and sublethal effects on larvae depending on the pH values. Pressing need of the aquaculture industry to have a user friendly and economically viable technology for the removal of ammonia, which can be easily integrated to the existing hatchery designs without any major changes or modifications. Only option available now is to have biological filters through which water can be circulated for the oxidation of ammonia to nitrate through nitrite by a group of chemolithotrophs known as nitrifying bacteria. Two types of bioreactors have been designed and developed. The first category named as in situ stringed bed suspended bioreactor(SBSBR) was designed for use in the larval rearing tanks to remove ammonia and nitrite during larval rearing on a continuous basis, and the other to be used for nitrifying freshly collected seawater and spent water named as ex situ packed bed bioreactior(PBBR). On employing the two reactors together , both penaeid and non-penaeid larval rearing systems can be made a closed recirculating system at least for a season. A survey of literature revealed that the in situ stringed bed suspended reactor developed here is unique in its design, fabrication and mode of application. |

Abstract: | Spike disease in sandal is generally diagnosed by the manifestation of external symptoms. Attempts have been made to detect the diseased plants by determining the length/breadth ratio of leaves (lyengar, 1961) and histochemical tests using Mann's stain (Parthasarathi et al., 1966), Dienes' stain (Ananthapadmanabha et a/., 1973) aniline blue and Hoechst 33258 (Ghosh et a/., 1985, Rangaswamy, 1995). But most of these techniques are insensitive, indirect detection methods leading to misinterpretation of results. Moreover, to identify disease resistant sandal trees, highly sensitive techniques are needed to detect the presence of the pathogen. In sandal forests, several host plants of sandal like Zizyphus oenop/ea (Fig. 1.3) also exhibit the yellows type disease symptoms. Immunological and molecular assays have to be developed to confirm the presence of sandal spike phytoplasma in such hosts. The major objectives of the present work includes:In situ detection of sandal spike phytoplasma by epifluorescence microscopy and scanning electron microscopy.,Purification of sandal spike phytoplasma and production of polyclonal antibodies.,Amino acid and total protein estimation of sandal spike phytoplasma.,Immunological detection of sandal spike phytoplasma., Molecular detection of sandal spike phytoplasma.,Screening for phytoplasma in host plants of spike disease affected sandal using immunological and molecular techniques. |

Abstract: | The most common and conventional method for removing turbidity from water is by coagulating with alum or iron salts, and settling the precipitate in suitably designed clarifiers followed by filtration. But the sludge produced is bulky, difficult to dewater and accumulates in the dumping grounds causing environmental problems. Synthetic polymers such as polyacrylamide and polyethyleneoxide have been investigated for their ability to remove turbidity. They overcome many of the disadvantages of conventional methods, but are cost—effective only when rapid flocculation and reduction in sludge volume are demanded. Considering the aforementioned situation, it was felt that more easily available and eco-friendly materials must be developed for removing turbidity from water. The results of our studies in this direction are presented in this thesis. The thesis comprises of nine chapters, with a common bibliography at the end. Chapter 1 gives an introduction to the nature of turbidity and colour usually present in water. Chapter 2 discusses the nature and availability of the principal material used in these studies, namely chitosan. Chapters 3 to 8, which deal with the actual experimental work, are further subdivided into (a) introduction, (b) materials and methods, (c) results and discussion and (d) conclusions. Chapter 9 summarises the entire work so as to put the results and conclusions into proper perspective. |

Abstract: | Loss of natural sandal populations due to illicit felling, forest encroachment and spike disease have an adverse effect on genetic diversity of the species. To initiate any genetic improvement programme in sandal, a precise understanding of the population genetic diversity structure is essential. The concern over the loss of genetic variability in sandal is particularly critical, as there is hardly any information regarding the diversity status of the natural populations. Identifying fast growing, disease resistant, oil rich sandal trees through breeding and their mass multiplication for afforestation are the best method for ensuring sustainable supply of superior sandalwood. The healthy sandal trees existing in heavily spike diseased area can be used as a promising starting point for any such breeding programme (Venkatesh, 1978). So far, no genetic information is available regarding the resistant nature of spike disease evaded trees left in heavily infected patches. The high rate of depletion of the superior trees in South Indian sandal reserves due to illegal felling and spike disease has necessitated an urgent need for conservation of the surviving trees.Widespread occurrence of spike disease in Marayoor forest reserve was reported in 1981 (Ghosh and Balasundaran, 1995). Because of the high density of trees and varying intensity of spike disease, Marayoor sandal population was found to be ideal for experimental studies in sandal (Ghosh et al., 1985). Fifteen trees of reserve 51 of Marayoor range had been selected as candidate plus trees for growth and spike disease evasion . These trees have been selected for mass multiplication through tissue culture technique. |

Abstract: | The Kerala Water Authority requested the School of Environmental Studies to carry out investigations on the mechanism of sporadic mobilization of iron and odour in the raw water drawn to the drinking water treatment plant. The currently used treatment process failed to remove iron completely. This led to problems in the filter and complaints of taste and colour due to iron in the finished water. The sporadic nature of the problem itself made the trouble shooting difficult. The problem was looked in from three points of view. 1. Influence of environmental (climatic) conditions on the dynamics of the relevant basin of the reservoir. 2. Influence of the physical dynamics on the physico — chemical quality of water. 3. Identification of cost-effective treatment processes to suit the existing plant. Since the problem emerged only during the post- monsoon to pre-monsoon months, a related problem was investigated, namely, influence of anions on the oxidation of Fe(II) in natural waters by air. This is presented in Part II of the dissertation. |

Abstract: | The tiniest Union territory of India, Lakshadweep, is an archipelago, with an area of 32 Sq. km. consisting of 12 atolls, three reefs and five submerged banks, lies between 8° and 12°30'N latitudes and 71° and 74" E longitudes. It is one of the most important and critical territories of India from economic and defence point of view. Specialised environment having typical geological set up, Lakshadweep is ecologically sensitive to even slight climatic or anthropogenic interference. Pollution of coastal seas, over exploitation and contamination of the fresh water sources are thus become great concerns to the existence of the island. Typical geological set up and interference cause threat to the ecology of the fragile environment and resources of the island as well as its resources. Marine pollution and ground water contamination are concerns in this regard. Even though attentions were made to assess the physico—chemical and bacteriological status of the marine and groundwater systems separately, an integrated approach has not been evolved. The present study with its broad objectives is attempted for an integrated assessment of microbiological, physicochemical and biological characteristics of the surrounding seawater and microbiological and physico—chemical characteristics of the ground water in Kavaratti island. Abstract: | This thesis Entitled Colour removal from dye house effluents using zero valent iron and fenton oxidation.Findings reported on kinetic profile during oxidation of dyes with Fenton’s reagent are in good agreement with observations of earlier workers on other organic substrates. This work goes a step further. Critical concentration of the dye at which the reaction mechanism undergoes transition has been identified.The oxidation of Reactive Yellow showed that the initial rates for decolorization increased linearly with an increase in hydrogen peroxide concentration over the range studied. Fenton oxidation of all dyes except Methylene Blue showed that the initial rates increased linearly with an in the ferrous sulphate concentration. This increase was observed only up to an optimum concentration beyond which further increase resulted in a decrease in the initial rates. Variation of initial rates with Ferrous sulphate concentration resulted in a linear plot passing through the origin indicating that the reaction is first order with respect to ferrous sulphate. |

Abstract: | The current water treatment technology is oriented towards the removal of contaminants, mostly organic compounds, by activated carbon. Activated carbons are classified as Granular Activated Carbons (GAC) and Powdered Activated Carbons (PAC) on the basis of the particle size of the carbon granules. Powdered carbons are generally less expensive than granular carbon, operating costs with powdered carbon could be lower. Though powdered activated carbon has many advantages over granular carbon, its application in large-scale separation process is limited by difficulty in recovery and regeneration. Deposition of magnetic iron oxide on carbon particles provides a convenient way of recovering the spent carbon from process water. The study deals with the preparation and physico-chemical characterization of magnetic iron oxide loaded activated carbons. The evaluation of absorption properties of magnetic iron oxide loaded activated carbon composites. The target molecules studied were phenol, p-nitro phenol and methylene blue. The feasibility of magnetic separation of iron oxide loaded activated carbons were studied and described in this thesis. |

Abstract: | Present work is aimed at development of an appropriate microbial technology for protection of larvae of macrobrachium rosenbergii from disease and to increase survival rate in hatcheries. Application of immunostimulants to activate the immune system of cultured animals against pathogen is the widely accepted alternative to antibiotics in aquaculture. The most important immunostimulant is glucan. Therefore a research programme entitled as extraction of glucan from Acremonium diospyri and its application in macrobrachium rosenbergii larval rearing system along with bacterians as microspheres. The main objectives of the study are development of aquaculture grade glucan from acremonium diospyri, microencapsulated drug delivery system for the larvae of M. rosenbergii and microencapsulated glucan with bacterian preparation for the enhanced production of M. rosenbergii in larval rearing system. Based on the results of field trials microencapsulated glucan with bacterin preparation, it is concluded that the microencapsulated preparation at a concentration of 25g per million larvae once in seven days will enhance the production and quality seed of M. rosenbergii. |

Abstract: | The main objective of the work undertaken here was to develop an appropriate microbial technology to protect the larvae of M.rosenbergii in hatchery from vibriosis. This technology precisely is consisted of a rapid detection system of vibrios and effective antagonistic probiotics for the management of vibrios. The present work was undertaken with the realizations that to stabilize the production process of commercial hatcheries an appropriate, comprehensive and fool proof technology is required primarily for the rapid detection of Vibrio and subsequently for its management. Nine species of Vibrio have been found to be associated with larvae of M. rosenbergii in hatchery. Haemolytic assay of the Vibrio and Aeromonas on prawn blood agar showed that all isolates of V. alginolyticus and Aeromonas sp., from moribund, necrotized larve were haemolytic and the isolates of V.cholerae, V.splendidus II, V.proteolyticus and V.fluvialis from the larvae obtained from apparently healthy larval rearing systems were non-haemolytic. Hydrolytic enzymes such as lipase, chitinase and gelatinase were widespread amongst the Vibrio and Aeromonas isolates. Dominance of V.alginolyticus among the isolates from necrotic larvae and the failure in isolating them from rearing water strongly suggest that they infect larvae and multiply in the larval body and cause mortality in the hatchery. The observation suggested that the isolate V. alginolyticus was a pathogen to the larvae of M.rosenbergii. To sum up, through this work, nine species of Vibrio and genus Aeromonas associated with M.rosenbergii larval rearing systems could be isolated and segregated based on the haemolytic activity and the antibodies (PA bs) for use in diagnosis or epidemiological studies could be produced, based on a virulent culture of V.alginolyticus. This could possibly replace the conventional biochemical tests for identification. As prophylaxis to vibriosis, four isolates of Micrococcus spp. and an isolate of Pseudomonas sp. could be obtained which could possibly be used as antagonistic probiotics in the larval rearing system of M.rosenbergii. |

Abstract: | The study conducted on the salinity intrusion and seasonal water quality variations in the tidal canals of cochin. The main objectives are, salinity intrusion profile, water quality variation of the surface water of the canals,hierarchical utility of the water bodies and to understand the non-conservative components in the water body. The parameters monitored werepH,temperature,alkalinity,conductivity,DO(dissolvedoxygen),COD(chemical oxygen demand),BOD(biochemical oxygen demand0,chloride, total hardness, calcium hardness, dissolved phosphate, nitrate, total iron, sulphate, turbidity, total coliform and SUVA at 254nm. The tidal canals of GCDA were found to be creeks extending to the interior, canals inter connecting parts of the estuary or canals with seasonally broken segments. Based on utility the canals could be classified as: canals heavely polluted and very saline,canals polluted by urban waste , canals having fresh water for most part of the year and not much polluted, fresh water bodies heavily polluted. During the rainy months carbon fixation by plankton is nonexistent,and during the dry months Chitrapuzha becomes a sink of phosphate. The study indicated abiotic subrouts for dissolved phosphate and revealed the potential pitfalls in LOICZ modeling exercise on sewage ladentidal canals. It was also found that all canals except for the canals of West cochin and chittoorpuzha have fresh water for some part of the year. The water quality index in the durable fresh water stretches was found to be of below average category. |
